Proximate, Elemental And In-vitro Anti-sickling Analysis Of Leave Extracts Of The Three Khaya Species Found In Nigeria.

Khaya with trade name African Mahogany is generally referred to as Oganwo in the Yoruba speaking tribe of Nigeria. Khaya belongs to the family Meliaceae and is widely distributed in Africa. However, only three species Khaya senegalensis (KS), Khaya grandifoliola (KG) and Khaya ivorensis (KI) exists in Nigeria along the Western Africa Region and they are phenotypically alike, hence very difficult to differentiate one from another. Traditionally in Nigeria, Traditional Medical Practitioners (TMPs) use Khaya to treat different arrays of diseases including Malaria, and Sickle cell disease. However, the three species are treated as same plants, hence the need for proper macroscopic and microscopic characterization of the species. Anti-sickling property of KS was earlier determined and was discovered to possess anti-sickling activity. Proximate and elemental analysis of the leaves and the anti-sickling efficacy of the leaf extracts of the three Khaya species (Khaya senegalensis (KS), Khaya grandifoliola (KG) and Khaya ivorensis (KI)) found in Nigeria were studied. The macroscopic and microscopic characteristics of the leaves were also studied. This is with the view to solve the identification problems. The proximate and elemental analysis was determined using the simple analytical procedure as described by the Association of Official Analytical Chemistry (AOAC) of 1987, 13th Edition Washington DC P1094 and Dry Ashing method of AOAC 2006 respectfully. Leaf anatomy were also done through leaf peeling and clearing to study the venation patterns, abaxial and adaxial features to solve the problems of identification. The anti-sickling properties of KS, KG, and KI studied using inhibitory and reversal anti-sickling in-vitro assay. The thin layer chromatographic fingerprints (TLC) of the most active extracts was also done to know and determine the different numbers of compound present in each extracts. The proximate analysis revealed that Khaya leaves are very rich in minerals, proteins and carbohydrates. The elemental analysis showed that Khaya leaves contains essential elements within the limits of dietary requirement. The leaf anatomy revealed some distinctive features that can be used in identifying each of the species studied from another.
