Comparative Genomics/Transcriptomics Valencia-D., Janice [1], Neubig, Kurt [2]. Evolutionary dynamics of orchid mitogenomes. Plant mitogenomes have more or less constant gene content but are highly variable in terms of non-coding DNA content, architecture and size. The presence of repeats that recombine frequently causes genomic reshuffling of big portions of the genome, and the acquisition of material through horizontal gene transfer provides new genetic material in the genome. However, there are 25 times fewer land plant mitogenomes than plastomes sequenced to date, and only three for orchids. Therefore, knowledge about the evolution of the mitogenome in land plants is limited. Here, we explore the evolution of the orchid mitogenome from a phylogenetic perspective. For this research, the mitogenomes were assembled from short paired-end reads obtained using Illumina sequencing technology. We developed a workflow to confidently recover mitochondrial sequences, even for regions without references in databases. We identified changes in genomic content through time and analyzed the effects of genomic rearrangements and gene gain/loss in orchid mitogenomes. Our results present new dynamics in orchid organellar genomes and shed light on a broader understanding of the evolution of angiosperm mitogenomes.
